Multiplex and ultra-sensitive immunoassays

NanoBiotec offers various cost-effective immunoassays to obtain maximum data from miniscule samples. Our immunoassays can be bead-based cytometric assays or glass slide-based antibody microarrays to quantify multiple analytes simultaneously in various samples. Samples can be serum, plasma, cell or tissue lysates, cell culture supernatants, milk, saliva, nasal lavage, exhaled breath condensate, urine, tear, etc. Variety of targets or biomarkers can be quantitated, such as cytokines, transcription factors, protein phosphorylation, histone acetylation, mRNA, miRNA, etc.  Our services include: 

Pre-defined Panels: Choose from hundreds of existing panels, which contain 1 to 80 targets or biomarkers involved in signal pathways of cancer, inflammation, autoimmunity, kidney toxicity, liver injury, metabolism, neurodegenerative diseases. Mix and Match Panels: Use the mix and match system if you are interested in ordering a subset of specificities from one of our pre-defined panels. Custom Panels: Design custom panels to combine analytes from multiple panels or request analytes that are not currently in our catalog. Each request will be evaluated for feasibility on a case-by-case basis.

    *  A fully automated, ultrasensitive, multiplexed immunoassay or Digital ELISA.  


 *  Simoa is based upon the isolation of individual immunocomplexes on paramagnetic beads using standard ELISA reagents. The main difference between Simoa and traditional immunoassays lies in the ability to trap single molecules in femtoliter-sized wells, allowing for a “digital” readout of each individual bead to determine if it is bound to the target analyte or not.


 *  Detection and quantification of low-abundance biomarkers difficult or impossible to measure with conventional assays. 


 *  Quantification of proteins as low as femtogram (fg)/ml in the samples


 *  The digital nature of the Simoa technology allows an average of 1000 x sensitivity increase over conventional ELISA assays with CVs < 10%.

  * We offer ELISA assays in various formats: direct or indirect, standard sandwichor Competitive ELISA, as well as ELISpot/ In-cell ELISA


 * Our ELISpot detects antigen-specific T cells in frequencies as low as 0.0001% within PBMC; that is, a single cytokine-secreting T cell can be reliably identified among one million bystander cells.


 * ELISpot is among the most useful for determining cell-mediated immunity because of its sensitivity and specificity in detecting rare antigen-specific T cells (or B cells). It provides qualitative information regarding the specific cytokine or other secreted immune molecule.


 * Our ELISpot is frequently utilized for immunogenicity testing, immune response monitoring, epitope discovery, and helper T cell response evaluation.

# Typically detect lower levels of protein with lower sample consumption than traditional methods. This can be particularly important when handling very limited volumes of precious samples from blood or tumor extracts. Traditional Western Blot

  •  Our traditional Western blot uses high-quality pre-cast gels with various chemistries (such as SDS-PAGE, Native-PAGE, etc.), as well as many well-validated specific antibodies, to obtain high resolution and consistent performance among multiple experiments.
  • LiCor Odyssey Near-Infrared Imaging technology is used to scan the probed target proteins giving a wider dynamic range, two-color (green and red) detection, and quantification with higher sensitivity when compared to common chemiluminescence methods.

Digi-Western Array

  • A high-throughput antibody-based microarray combining SDS-PAGE based protein separation with immobilization on Luminex beads.
  • The combination of MW resolution, sensitivity, and signal linearity on the Luminex platform enables the rapid quantification of hundreds of specific proteins and protein modifications in complex samples.
  • Parallel profiling of currently up to 600 total and phosphoproteins in just 50 ug of samples (cells, tissues, …) to support biomarker discovery pathway mapping, drug MoA studies, lead compound characterization, etc.

In-Cell Western Blot (In-Cell ELISA™)

  • Our assay combines the specificity of Western blot with the reproducibility and throughput of ELISA, which is a simple and cost-effective means for quantification of intracellular signaling in whole cells.
  • This assay involves seeding cells in 96- or 384-well microtiter plates followed by fixation/permeabilization and subsequent labeling with activation state-specific or control antibodies, infrared-conjugated secondary antibodies, and/or far-red DNA dye.
  • The well-level data acquired by the Odyssey® Imaging System permits accurate measurement of signal induction or inhibition, with the potential for multiple treatments, endpoints, and replicates on each plate.
  • Our assays quickly and accurately measure relative protein levels in many samples, detect proteins in situ in a relevant cellular context.
  • This is used for high-throughput analysis of protein phosphorylation (effects of drug compounds on signaling pathways; timing/kinetics of signal transduction), caspase-3 activation, cell surface proteins, and receptor internalization, RNAi library screens, cell proliferation, etc.
